Describe each of the following, as well as some of their usesin the laboratory:

    1. polyvinylchloride (PVC)

    2. polytetrafluoroethylene

    General Laboratory

    1. Polyvinylchloride,or PVC, is a type of plastic. Polyvinylchloride is clear, can either beflexible or rigid, and it can also be either non-autoclavable or autoclavable.Flexible, autoclavable polyvinylchloride is used to make plastic tubing for usein the laboratory. This tubing can be used in various pieces of automatedlaboratory equipment, including continuous flow analyzers. Polyvinylchloridethat is rigid and non-autoclavable is used to make plastic bottles forlaboratory use.

    2. Polytetrafluoroethyleneis a compound that sometimes goes by its trademark, Teflon. Teflon is acompound that has a slippery surface, and it is tough and stable over a widerange of temperatures It also has a non-wettable surface. In the laboratory,Teflon is used to make transfer disks for centrifugal analyzers, and it is alsoused to make stoppers and gaskets.

Describe each of the following, as well as some of their usesin the laboratory:

    1. polypropylene

    2. polycarbonate

    General Laboratory

    1. Polypropyleneis a type of plastic. Polypropylene is rigid, translucent, and it has anon-wettable surface, similar to Teflon. Polypropylene can also be autoclaved, because it has a melting point of 320 degrees Fahrenheit (160 degrees Celsius). This high melting point makes polypropylene able to withstand the hightemperatures of an autoclave. In the laboratory, most pipette tips are made outof polypropylene, as are some test tubes and diluent trays.

    2. Polycarbonateis another type of plastic. Sometimes, polycarbonate goes by the trademarkname, Nalgene. Polycarbonate plastics are flexible, lightweight, and strong. They are able to withstand high impacts without breaking upon impact. In thelaboratory, they can be used as a substitute for glass, because of theirshatter-resistant capabilities.

Define desiccant and list some desiccants that may be used inthe laboratory.

    General Laboratory

A desiccant is a material that readily absorbs water(hygroscopic). Desiccants are used to keep things dry and to prevent them frombecoming hydrated or in contact with water. In the laboratory, some things thatdesiccants are used to keep dry are reagents, gases used in gas chromatography, and thin-layer chromatography (TLC) plates. Usually, a desiccator is used. Adesiccator is typically made out of glass, and the material in question isplaced on a small shelf, with the desiccant placed below the shelf. A glass lidmakes an air tight seal on the vessel. Some of the materials that can be usedas desiccants include silica gel, calcium sulfate, sodium hydroxide, andmagnesium perchloride. One of the least effective desiccants is silica gel, andthe most effective desiccant is magnesium perchloride.

Briefly describe Kimax and Corex glass.

    General Laboratory

Kimax is a brand name for borosilicate glass. Borosilicateglasses contain at least 5% boric acid in their composition. They are also madeup of sodium oxide, potassium oxide, calcium oxide, and silica. Borosilicateglasses are very heat resistant and strong as well. They also have a lowthermal expansion coefficient, meaning that they do not expand much whenheated. In the laboratory, flasks, beakers, and pipettes are usually made ofborosilicate glass. Pyrex is another brand name of borosilicate glass products.

Corex is a brand name for aluminum-silicate glass.Aluminum-silicate glass is even stronger than borosilicate glass -- about sixtimes stronger. Aluminum silicate glass also exhibits resistance to scratchingand etching from alkaline materials. Corex glass is used in the laboratory inthe form of thermometers and centrifuge tubes.

Discuss laboratory safety: World Health Organization (WHO)rules.

    General Laboratory

Each laboratory must have a manual that contains safetyregulations. Labs should have sufficient workspace and accommodate disabledworkers. Areas with patient access should be separate from the work area. TheWHO has established rules for laboratory safety:

The laboratory must be separate from areas of unrestrictedtraffic flow.

Entry to the laboratory should be through double doors or avestibule.

All surfaces should be water-resistant for easy cleaning and wellsealed, impervious to water, and resistant to chemicals, such as acids andsolvents.

The laboratory itself must be sealable and allow for gaseousdecontamination.

Windows must be kept closed and sealed, and must be resistant tobreakage.

Airflow must be into the laboratory from access rooms.

Negative pressure airflow must be maintained and intake andoutput filtered through a HEPA filter. Output must be discharged outside of thebuilding and at a distance from air intakes.

The water supply must be outfitted with anti-backflow devices.

All effluent must be decontaminated before discharged into thesewer system.

Biological safety cabinets must have a negative airflow filter.

    Briefly discuss universal precautions.

    General Laboratory

Universal precautions are used in the laboratory and othermedical settings to help protect healthcare workers from diseases and poisonstransmitted via bodily fluids and secretions (blood, semen, amniotic fluid, vaginal secretions, sweat, tears, saliva, urine, feces, wound drainage andCSF). Universal precautions became required practice after the outbreak of AIDSin 1980. Universal precautions means washing thoroughly and wearing protectivegear, such as gloves, goggles or glasses, face shields, shoe covers, gowns, andanything else deemed appropriate by Infection Control. Which precautions needto be practiced in a given situation depend on the possibility of a healthcareworker coming in contact with the bodily fluids or secretions, not on thepatients known health status. Assume all patients are always infectious.

Briefly describe some of the aspects involved in the properstorage of chemicals in the laboratory.

    General Laboratory

Proper storage of chemicals in the laboratory is vital tomaintaining technician safety and a safe work place. All lab chemicals musthave the proper labels and be stored in the proper container. For example, materials that are flammable need to be stored in a fire safety cabinet, tominimize the risk of fire. Solvents, on the other hand, need to be stored in arefrigerator that is explosion-proof. In addition to using proper storagecontainers and equipment, chemicals that are incompatible with each othershould never be stored together. An example of this would be storing oxidizingagents next to reducing agents. Therefore, chemicals should never be storedalphabetically, as this may place incompatible chemicals next to each other ina storage situation. The amount of flammable substances and solvents that arestored should be limited, further reducing the risk of fire. Store largecontainers on a low shelf, close to the ground, to minimize dangers if thesecontainers break. Check storage areas monthly to make sure that everything isstored properly.
